| The instrument is mainly used to identify proteins by the so-called “peptide mass fingerprint” approach. Briefly, proteins visible in Coomassie-stained bands in an SDS-PAGE are digested within the gel with a specific endoproteinase. Peptides are extracted and their molecular weights are determined in the MALDI-ToF. The measured molecular weights of the peptides are compared with the calculated molecular masses of peptides derived from all proteins in a database after digestion in silico. The instrument has the advantage that samples can be analysed without any time-consuming desalting procedure, by using the thin-layer technique for the preparation of the MALDI matrix on the sample plate (“target”). | ||||
