FILM Microscopes

Four generations of confocal laser scanning microscopes:

Leica TCS SPE, providing excitation wavelengths of 488, 561 and 635 nm. In addition it has a widefield UV illumination. This system comes with only one PMT detector. Multicolor imaging is thus only possible in sequential mode using the ultra high dynamic PMT.

Leica TCS SP5, providing excitation wavelengths of 458, 476, 488, 496, 514, 561 and 633 nm. In addition it has an UV diode laser with excitation at 405 nm. Besides the detection system for 4 fluorescence channels simultaneously and one transmission channel, this systems is also equipped with an acusto-optical-beam-splitter which provides the highest flexibility for setting up the required experimental parameters. This system enables confocal and multiphoton imaging.

Leica TCS SP5 (STED), providing excitation wavelengths of 458, 476, 488, 496, 514, (561) and 633 nm. The system lacks an UV excitation source. The detection system is equipped with for 4 fluorescence channels and one transmission channel. Be aware that the STED depletion laser is removed (no STED, only confocal imaging).

Leica TCS SP8, providing excitation wavelengths of 458, 476, 488, 496, 514, 561 and 633 nm. In addition it has an UV diode laser with excitation at 405 nm. Besides the detection system for 4 fluorescence channels simultaneously, this systems is  equipped with an acusto-optical-beam-splitter which provides the highest flexibility for setting up the required experimental parameters. It  provides two new Leica HyD hybrid detectors for brilliant imaging with super-sensitivity and contrast and two PMTs.

One super-resolution STED microscope:

Abberior Instruments STED 775 & 595 QUAD Scanning, 3D, providing a multicolor confocal scanning system with two super-resolution STED channels. The system is based on an Olympus IX83 microscope (4-color LED illumination source and a monochrome widefield camera) equipped with the Abberior QUAD scanner (Scan field: >80×80 μm, Line frequency: 2 kHz). The STED system has two pulsed excitation laser sources with wavelengths at 561 and 640 nm and a pulsed STED laser at 775 nm.

For 595 STED the excitation is 485 nm for GFP/YFP-like dyes.

And one motorized research microscope: 

Leica DM 6000B, provides brightfield, differential interference, polarization and fluorescence contrast. It is equipped with a 1.4-megapixel, 12-bit monochrome CCD camera.
